Efficacy of Cetuximab and 4-PBA Combination Therapy in Human Oral Squamous Cell Carcinoma Cells

Cetuximab is a powerful anti-neoplastic agent that can inhibit cell growth in oral squamous carcinomas (OSCCs). Unfortunately, there are cases with unfavorable outcomes. Because few studies have focused on the combined effects of cetuximab and histone deacetylase (HDAC) inhibitors, we aimed to evaluate antitumor effect combination HDAC inhibitors human OSCC lines, investigate mechanism apoptosis enhancing activity thereof. We used lines treated several inhibitors. The WST assay ApoToxGlo™ Triplex Assay determined survival. employed TdTmediated dUTP-biotin nick end labeling method detect apoptosis. western blotting examine acetylation status, ER stress markers, epidermal factor receptor (EGFR) signaling pathways. Our results show 4-phenyl butyric acid (4-PBA) remarkably decreases vitro. In addition, treatment resulted increased EGFR mRNA expression promoted activation ERK. induced at significantly higher frequency than did either alone. conclusion, 4-PBA more effective against cells, allowing for potential clinical applicability therapies.
